As far as I know PHPMailer should work on any system that has PHP installed.

PHPMailer works great with any SMTP outgoing mail server. If your Exchange server has SMTP set up, PHPMailer will work with it.

For help configuring your Exchange server, try the Windows forum (not sure if this would qualify as an IIS issue).
