I'm not entirely sure this is possible. What are you trying to do?

        hummm SOUNDS LIKE A HACK...

well i shouldn't but im gonna tell u anyway since i achieved it myself... (for educational purposes only)

        1) u'll need a U3 enabled usb drive (an old one wont work)
        2) u'll need the u3 customizer program (just google it)
        3) create an image of the program with the edited autorun.inf (has 2 be less than 6MB)
        4) edit u3 device w new iso image of program (always backup U BEEN WARNED)
        5) the device is ready to be plugged and the program stated in the autorun.inf will run automatically

u can use autoplay (not same as autorun) with same autorun.inf file and it will work but it will need some clever social engineering and user interaction...

        enjoy

with a normal USB drive, you can write an autorun.inf, but that will at most pop up a window with the option. Since it's an exe, if you want to run it on someone elses computer automatically that's probably illegal, but on your own computer, you can change a registry key to enable USB drives to autorun.
