Well you need to make sure that the Server that's hosting your application has the same .NET Framework installed that you developed your website with. So if you used Visual Studio 2008 this Framework could be 2.0, 3.0. or 3.5....
You also need to make sure that any libraries that your website referenced are installed on the server. For example, if you used the Ajax Toolkit then you will have to have that installed on the server because it has server components (whereas JQuery does not have any server components and does not need to be "installed"...b ut does need to be part of your application). Another example would be if your application uses Word for creating a .doc file...Word would have to be installed on the server (or at least the redistributable class library that has the Word library included).
If your website uses a Database ... you need to make sure that it can connect to that database (this may mean that you have to set it up on the web server)
Once you're done setting up the web server you would Publish your website...and take the files that were generated by the Publish and move them to your sever.
